Can I safely reformat my HD-PE500U2 external USB drive?

The drive is currently FAT32 and I want to use Windows Backup on it. Checked the online documents and can't find any reference to reformatting as NTFS. Will this cause any problems? The drive will only be used with Windows servers as USB external drive.

The drive can handle NTFS just fine, but any data or programs you have on the drive will be wiped off in a format. So back up any data first.
