I've got a new Buffalo HD-CE10TU2B which I've just finished putting all of my movies and music on.  It still has about 400gb free but this morning it's just started beeping and clicking like a clock - beep, click, beep, click...  It wont show up at all in My Computer as a drive at all but I hear the usb notification on my laptop when I plug it in.  It's like it's completely kicked the bucket.

Is there anything I can do?

I cant run a recovery program because it does not show up as a drive in My Computer at all...

Does it show up in disk management?  It sounds like a bad HDD, I would contact support for a replacement.
