RAID is not a replacement for a backup! Here's why.
I've checked that the HD is NFTS formatted.
And I've been using this as my backup drive with lots of data files already.
I wonder if I change it from NTFS to FAT32, will my data files be affected?

If you format the HDD all your data on it will of course be erased.
